Intended Use:
This product is used for the in vitro quantitative determination of free thyroxine (FT4) in human serum. Tetraiodothyronine (T4) is a thyroid hormone synthesized and secreted by thyroid follicular epithelial cells. The formation and release of T4 is regulated by thyroid stimulating hormone (TSH). The majority of T4 in the body (over 99%) is bound to transport proteins, including thyroid binding protein (TBG), prealbumin (TBPA) and albumin, with only a very small amount (approximately 0.04%) of T4 present in the free form in the serum. T4 affects the metabolism of proteins, lipids and carbohydrates in the body, and in peripheral tissues, it is further metabolized by 5'deiodination to more active thyroid hormone (T3). Serum FT4 concentrations are highly correlated with the effective thyroxine ratio (ETR). FT4 is elevated in hyperthyroidism, medically induced or toxic hyperthyroidism, and elevated TBG from various causes, such as estrogen and hepatitis, also elevates T4 but not FT4; in cases of hypothyroidism from various causes, FT4 is significantly below the normal range. It is generally accepted that in clinical diagnosis, FT4 measurement has the advantage of being independent of changes in the concentration and binding characteristics of its conjugated proteins and best reflects thyroid function, and the combination of FT4 with TSH improves the diagnostic accuracy.
