For quantitative determination of ferritin in human serum.
Principle:
The latex particles coated with anti human ferritin are agglutinated when they react with samples that contain ferritin. The latex particles agglutination is proportional to the concentration of the ferritin in the sample and can be measured by turbidimetry.
REAGENTS
R1 (DILUENT): Glycine buffer 20 mmol/L, pH 8.5
R2 (LATEX): Latex particles coted with polyclonal anti human ferritin. pH 8.2
CALIBRATION (Optional)
Serum Ferritin Calibrator is recommended.
Ferritin concentration is stated on the label vial.
PRECAUTIONS
1. The reagents contain sodium azide 0.95 g/L.. Avoid any contact with skin or mucous.
2. The reagents from human donors have been given negative results to anti HIV ½, HbsAg and anti HCV . Handle cautiously is recommended.
REAGENT PREPARATION
R1 :Ready to use.
R2 :Ready to use. Shake gently the vial before use.
INTERFERNCES
Bilirubin (20 mg/dL), hemoglobin (10 g/L), rheumatoid factor (600 IU/mL), do not interfere.
Lipemia interferes.
ADDITIONAL EQUIPMENT
Spectrophotometer or photometer thermostable at 37˚C with a (540±20) nm filter.
PROCEDURE
Wavelength : 540 nm
(530-550 nm) Working temperature : 37°C
Optical path : 1 cm
Assay type : Turbidimetry
Direction : Increasing
1. Bring the reagents and the photometer (cuvette holder) to 37°C .
2. Adjust the instrument to zero with distilled water.
3. Pipette into a test tube
4. Mix and pipette R2 Reagent into cuvette
5. Mix well and record the absorbance immediately (Abs.1) and after 5 minutes of the reagent R2 addition read the final absorbance (Abs. 2)
