METHOD HISTORY
5-BR-PAPS use in routine zinc determinations because, this compound is water soluble, and has a higher molar absorptivity. Results by the proposed method developed with 5-Br-PAPS correlated well with those by atomic absorption spectrophotometry. This method is fast,sensitive and linear over the entire concentration range of zinc in human serum, plasma or urine.
PRINCIPLE
Zinc produces with the specific complexant 5-Br-PAPS [(2-5-bromo-2- pyridylazo)-5-(N-propyl-N-sulfo-propylamino) phenol] a stable coloured complex, which colour intensity is proportional to the amount of zinc in the sample. The interference due do the oligoelements (iron and copper) present in the sample, are eliminated using peculiar reaction conditions and specific masking agents.
PRECAUTIONS
1. The reagents contain Sodium azide.
2. Handle with care. The disposal of the residues has to be made according to legal local regulations.
3. To avoid ingestion and contact with eyes,skin and mucous membranes.
SPECIMEN COLLECTION AND STORAGE
1. Serum or nonhemolyzed plasma:Use only heparin salts as anticoagulant.
2. 24 hour urine : Diluted 1:10
3. Shake and bring the samples at room temperature (15 – 25ºC) before using.
4. Turbid and icteric sera can interfere in the reaction raising values.
INTERFERENCES
EDTA anticoagulant masks zinc to 5-Br-PAPS chromogenic system. the test is not affected by the presence of conjugated and non-conjugated bilirubin up to 15 mg/dL, hemoglobin up to 0.5 g/dL and triglycerides up to1000 mg/dL,
