For the quantitative determination of iron in serum. METHOD HISTORY Iron measurements are used in the diagnosis and treatment of diseases and conditions such as iron deficiency anemia, hemochromatosis (characterized by a progressive increase in iron stores leading to organ impairment), chronic inflammatory disorders, hepatitis, and lead poisoning . Iron exists in the body as a component of hemoglobin and myoglobin as well as bound to transferrin for the transport in plasma and stored in ferritin. Principle: In an acidic medium transferrin bound iron dissociates into ferric ions Reduced iron reacts with the Ferene to form a blue complex which absorbs at 595 nm. The absorbance is directly proportional to the serum iron concentration. Precautions 1. All reagents are toxic. Do not pipette by mouth. Avoid all contact. 2. This reagent is for in vitro diagnostic use only. 3. Use only disposable material to avoid iron contamination. Rinse glass material with diluted HCl and copious dist. water. INTERFERENCES Bilirubin: No interference up to 10 mg/dL. Lipemia: No interference in the presence of triglycerides up to 1000 mg/dL. Ascorbic acid: No interference up to 20 mg/dL. MATERIALS REQUIRED BUT NOT PROVIDED 1. Accurate pipetting devices 2. Test tubes/rack 3. Timer 4. Spectrophotometer able to read at 595 nm. 5. Iron-free deionized water. 6. Heating bath/block (37°C). 1. Mix, read the absorbance against blank in 1 - 5 minutes (Abs.1) 2. Mix and place all tubes in heating bath at 37°C for 10 minutes. 3. After 10 minutes, read absorbance against blank.(Abs.2) 4. ΔAbs.= (Abs.2 – Abs.1)
