Combined method for highlighting Helicobacter pylori and epithelial mucins on sections
of gastric tissue; recommended section thickness 5 microns.
Product for the preparation of cyto-histological samples for optical microscopy.
Integrated method to show Helicobacter pylori and acid mucins in gastric tissue sections. Suitable section thickness 5 μm.
PRINCIPLE
Gastric epithelial mucins are oxidised by periodic acid and stain with Alcian Yellow (they normally unstained by the Alcian dyes).
They form an insoluble compound that will not be affected by the second part of the procedure when toluidine blue solution will stain bacteria.
METHOD
1) Bring section to distilled water.
2) Put on the section 10 drops of reagent A: leave to act 10 minutes.
3) Wash well in distilled water.
4) Put on the section 10 drops of reagent B: leave to act 5 minutes.
5) Wash in running water 2 minutes.
6) Put on the section 10 drops of reagent C: leave to act 5 minutes
7) Wash well in distilled water.
8) Put on the section 8 drops of reagent D and 2 drops of reagent E: leave to act 3 minutes.
9) Wash well in distilled water.
10) Dry the slide in the air
11) Dehydrate in alcohol; clear in xylene and mount.
