Application
Removal of glycogen from
- Hepatic tissue, paraffin-embedded sections: digestion on a histological section with a
solution of amylase is indicated when you want to remove the glycogen so as to observe
only the neutral epithelial mucins. It is the method of choice in liver biopsy.
- Muscle tissue: the examination of adjacent cryostat sections, one of which has been
treated with amylase, allows qualitative evaluation of the presence of glycogen.
Result
The removal of glycogen can be detected, after PAS reaction, by comparing the sample
section with an adjacent section of the same preparation that has not been treated with
amylase.

Removal of glycogen from tissue sections:
- Liver, paraffin embedded sections: the removal of glycogen is advisable in order to observe only epithelial neutral mucins.
This method is recommended for liver biopsy.
- Muscle frozen section.
METHOD
1) Bring sections to distilled water.
2) Bring the Amylase solution at room temperature.
3) Cover sections with amylase solution: incubate 10 minutes at room temperature.
4) Rinse the slide in distilled water several times.
5) Stain with P.A.S. reaction
