Application
Digestion on a histological section with a solution of diastase is always indicated when
you want to remove the glycogen so as to observe only the neutral epithelial mucins. It is
the method of choice in liver biopsy.
Result
The removal of glycogen can be detected, after PAS reaction, by comparing the sample
section with an adjacent section of the same preparation that has not been treated with
diastase.

PRINCIPLE
The Diastase essentially hydrolyses starch and glycogen on the sections of tissue. Usually digestion of glycogen is not complété on tissue fixed because of particular glycosidic bond that they are formed during conventional fixation and it is possible to observe a granular glycogen precipitate to a pôle of the hépatocytes.
