PrincipleBilirubin is the iron-free derivative of biliverdin, produced by haemoglobin degradation by the reticulo‑endothelial system. The Fouchet reaction involves strong oxidation‑reduction of the complex and conversion to biliverdin, producing characteristic colour changes. The reaction occurs mainly in bone marrow, spleen and liver. Bilirubin is insoluble in water and aqueous fixatives and may turn green after prolonged formalin fixation.
Method- 1) Hydrate sections in distilled water.
- 2) Apply 5 drops of reagent A to the section and add 5 drops of reagent B; incubate 5 minutes.
- 3) Rinse in distilled water.
- 4) Apply 10 drops of reagent C to the section; incubate 7 minutes.
- 5) Drain the slide without rinsing and dry first with filter paper then air‑dry 5 minutes.
- 6) Absolute alcohol 15 seconds, xylene and mount with balsam.
