PRINCIPLE
In this method, some components of connective tissue are selectively stained according to the different degree of affinity
between dyes and tissue macromolecules. In particular, connective tissue is markedly acidophil because of the presence of many
basic groups; therefore, it shows a strong affinity to acid dyes (picric acid and orange G) and a medium affinity to weak basic
amphoteric dyes (acid fuchsin and ponceau xylidine). A central role is played by phosphomolybdic acid that acts as a bridge
between tissue structures it attaches to (collagen fibrils, cell membranes) and aniline blue, a markedly basic amphoteric dye.
