PRINCIPLE
Sirius red picrate solution performs a double function:
- to stain collagen by the binding between Sirius red and some proteins containing basic amino acids
- to stain bilirubin by conjugation by picric acid
Method
1) Bring the section to the distilled water.
2) Dispense 10 drops of reagent A onto the section: leave to act for 50 minutes.
3) Wash briefly in distilled water.
4) Dispense 10 drops of reagent B onto the section: leave to act for 2 minutes. Repeat
twice.
5) Wash briefly in distilled water and drain the slide.
6) Dispense 10 drops of reagent C onto the section: leave to act for 3 minutes.
7) Leave to develop in tap water: 3 minutes.
8) Wash in distilled water and dehydrate rapidly by means of the ascending series of
alcohols, leaving for 1 minute in the last absolute: xylene and balsam.
