NAD/NADH quantification assay kit is based on several reactions involving various chemical compounds. The concentration of NAD+ is calculated through its activity as a cofactor of the reaction of the enzyme alcohol dehydrogenase (ADH) with ethanol. In a situation of maximum enzyme-substrate saturation, the rate depends solely on the concentration of available NAD+, which is transformed in its reduced form NADH.
The measurement of absorbance is carried out thanks to the ability of NADH to cede an electron to an intermediate electron donor: 1-methoxyPMS. This intermediate donor will cede the electron to MTT which will turn in formazan (from yellow to blue), absorbing at a wavelength of 570 nm.
