Staining Principle
N, N-dimethyl-m-phenylenediamine dihydrochloride and N, N-dimethyl-p-phenylenediamine dihydrochloride are both ammonium salts, both of which are positively charged after dissociation, diammonium. This ammonium salt is combined with the sulfated acid mucus substance to form a composites that has chromogenic result. and the reaction is very slow. It is necessary to add ferric chloride as a catalyst. On the one hand, the diammonium salt is oxidized to form a brown-black cationic chromogen, which speeds up the staining. On the other hand, the pH of the stain is reduced by 1.4. At this pH, the carboxyl groups on the slices cannot combine with the diammonium salt but only the sulfate group reacts with the diammonium salt to form a purple brown to brownish black complex. Then the carboxylated sialic acid mucus is stained blue by Aixian Blue (pH 2.5), so that the two main groups of acid mucus substances were shown separately.
