Reagent B is a weak alkaline aqueous solution of surfactant and inorganic acid. The pH value is in the range of 8.00-9.00. Reagent B is used for neutralization of weak acidic solutions which is of particular importance for DNA quantification.
It is used in conjunction with Reagent A100 to raise the sample’s pH value and avoid DNA degradation as well as improve the binding efficiency of DAPI.
Reagent B facilitates precise DNA content quantification of acidic cell samples by raising the pH.
Neutralization of acidic cell samples is important for DNA stabilization, and furthermore potentiates the fluorescence signals of various dyes, such as Propidium Iodide (PI). Please refer to the specific Application Notes for detailed instructions.
