Reagent Y200 is a lysis buffer that permeabilizes yeast cells, facilitating staining of the total cell population. Reagent Y200 is a neutral aqueous solution containing surfactants and inorganic salts. The pH value is between 6.9 and 7.1.
Y200 is developed for the Yeast Cell Count and Viability Assay using the NucleoCounter® NC-202™ automated cell counter. Reagent Y200 permeabilizes the yeast cell wall and membranes, allowing homogenous staining of the nuclei using DNA binding dyes such as acridine orange, DAPI and propidium iodide. Reagent Y200 also dilutes the yeast sample before counting. Please refer to the specific Application Notes for detailed instructions.
Y200 is developed as a Triton X-100-free alternative to Reagent Y100.
