Solution 2 – Propidium Iodide (PI) stains nucleic acid of fixed and permeabilized cells. The solution contains PI (20 µg/ml ∼ 30 µM ∼ 0.002%) and Triton X-100 (1 mg/ml ∼ 1.6 mM ∼ 0.1%) in PBS (pH 7.4). Solution 2 – PI can be used to quantify DNA content of e.g. animal cells and yeast cells, and is a competent dye for measurement of G0/G1, S and G2/M cell cycle phases. The fluorescence intensity of PI stained cell is in stoichiometric relationship to the DNA content. For accurate DNA content measurements using Solution 2 – PI, cells must be fixed/permeabilized and RNase treated prior to staining.
