Solution 12 – DAPI is a staining reagent containing 4’,6-diamidino-2-phenylindole (DAPI), which stains nucleic acids of fixed and permeabilized cells. Solution 12 – DAPI can be used to quantify the DNA content of e.g. animal cells to determine G0/G1, S and G2/M cell cycle phases. Solution 12 – DAPI is a competent dye for measurement of the cell cycle stages. The fluorescence intensity of DAPI stained cells is in stoichiometric relationship to the DNA content. For accurate DNA content measurements, cells must be fixed/permeabilized prior to staining. Solution 12 – DAPI is used in conjunction with Solution 10 and Solution 11.
