Mu-MLV (Moloney Murine Leukemia Virus) generates complementary DNA strand starting preferably from single-stranded RNA in the presence of a primer, or from RNA-DNA hybrid. Mu-MLV reverse transcriptase has lower RNase H activity than AMV reverse transcriptase, which is an advantage when synthesizing cDNAs from long mRNAs. Mu-MLV lacks the 3’ ’ 5’ exonuclease activity. It is recommended to use 10 U/µg RNA for optimum reverse transcription.
Applications
This enzyme is particularly suited for long cDNA fragments in the context e.g. of generating cDNA libraries or for first strand cDNA synthesis prior to RT-PCR.
