Dulbecco’s Modified Eagle Medium (DMEM) with 4500 mg/L D-glucose, 25 mM HEPES buffer, L-glutamine, and sodium pyruvate. What is DMEM? Dulbecco’s Modified Eagle’s Medium (DMEM), a modification of Eagle’s BME, is widely used for a range of mammalian cell culture applications. In comparison with BME, it contains a four-fold higher concentration of amino acids, vitamins, and additional components. A variety of DMEM versions are available in high glucose and low glucose formulations. DMEM requires supplementation with serum, typically 10% Fetal Bovine Serum (FBS), since this medium does not contain proteins, lipids, or growth factors. Each lot of DMEM is prepared from powdered base medium, tissue culture-grade water, and is sterile filtered using a 0.2 micron filter. Representative samples of each lot of DMEM are tested to confirm the absence of bacterial or fungal contamination using methods adapted from the current U.S. Pharmacopeia.
