SARS-CoV-2 is an enveloped positive-sense RNA virus that causes COVID-19. The spike (S) spike protein is located on viral membrane and gives the name of coronavirus family via resembling a crown-like structure. Main function of S-protein is the binding to recognition protein, ACE2, on host cells and facilitation of viral and cell membranes. S-protein is composed of two domains: S1 and S2 domains. S1 domain contains a vital subdomain for ACE2 binding namely, receptor binding domain (RBD) and N-terminal domain. Neutralization antibodies are mostly targeted to S1 domain rather than S2 domain. Also, S1 domain is less conserved part of Spike proteins among human coronaviruses.
