The ThruPLEX DNA-Seq Kit enables generation of high-quality DNA libraries from as little as 50 pg of input material, and features a user-friendly protocol that is as simple as any NGS library prep solution currently available on the market. This kit can be used to process fragmented double-stranded DNA (dsDNA) from any sample source, including genomic DNA isolated from cells/tissues, cell-free DNA, FFPE DNA, outputs from ChIP or CUT&RUN approaches, and RNA-derived cDNA.
The entire ThruPLEX DNA-Seq Kit workflow is performed in a single tube or well in about two hours and requires no intermediate purification steps or sample transfers that pose the risk of sample loss and reduced library complexity. ThruPLEX technology enables efficient repair and capture of dsDNA fragments as small as 50 bp and has been successfully paired with commonly used target enrichment platforms, allowing for a wide range of NGS-based applications. Compatible indexing reagents are described below and allow for multiplexing of up to 384 samples per sequencing run.
Unique Dual Index kits (plate format)
Unique Dual Index kits are designed for use with the vast majority of NGS library preparation kits from Takara Bio for multiplexed sequencing on Illumina sequencers. These kits contain indexed PCR primers that include the same adapter as the IDT for Illumina-TruSeq DNA and RNA UD Indexes, with Cat. # 634752 and 634756 carrying the “IDT for Illumina” indexes and Cat. # 634753–634755 carrying indexes that have been developed and validated by Takara Bio. These kits offer a total of 384 dual indexes for multiplexing of up to 384 samples.
